Remains of Boston Marathon bombing suspect claimed

BOSTON (AP) ? The body of Boston Marathon bombing suspect Tamerlan Tsarnaev, who was the subject of a massive manhunt and died after a gunbattle with police, was claimed on Thursday.

Department of Public Safety spokesman Terrel Harris said a funeral home retained by Tsarnaev's family picked up the 26-year-old's remains. He said he had no more information about plans for the remains.

The medical examiner determined Tsarnaev's cause of death on Monday, but officials said it wouldn't become public until his remains were released and a death certificate was filed. It was unclear on Thursday evening whether the death certificate had been filed.

Tsarnaev's widow, Katherine Russell, who has been living with her parents in North Kingstown, R.I., learned this week that the medical examiner was ready to release his body and wanted it released to his side of the family, her attorney Amato DeLuca said days ago.

Tsarnaev's uncle Ruslan Tsarni, of Maryland, said Tuesday night the family would take the body.

"Of course, family members will take possession of the body," Tsarni said. "We'll do it. We will do it. A family is a family."

After the hearse believed to be carrying Tsarnaev's body departed Boston, television stations reported that their helicopters followed it to the Dyer Lake Funeral Home in North Attleboro. About 20 protesters gathered outside the funeral home. An Associated Press photographer later saw a hearse leaving the home escorted by two police cars.

Dyer-Lake Funeral Director Tim Nye told The Sun-Chronicle newspaper late Thursday that the body was only brought to his funeral home temporarily and was transported to another facility, but didn't say where.

Tsarnaev, who had appeared in surveillance photos wearing a black cap and was identified as Suspect No. 1, died days after the bombing.

The April 15 bombing, using pressure cookers packed with explosives, nails, ball bearings and metal shards near the marathon's finish line, killed three people and injured more than 260 others. Authorities said Tsarnaev and his younger brother later killed a Massachusetts Institute of Technology campus police officer and carjacked a driver, who escaped.

Authorities said that during the gunbattle with police, the Tsarnaev brothers, ethnic Chechens from Russia who came to the United States about a decade ago, set off another pressure cooker bomb and tossed grenades before the older brother ran out of ammunition.

Police said they tackled the older brother and began to handcuff him but had to dive out of the way at the last second when the younger brother, 19-year-old Dzhokhar Tsarnaev, drove a stolen car at them. They said the younger brother ran over his brother's body as he drove away from the scene to escape.

Dzhokhar Tsarnaev was captured later, wounded and bloody, hiding in a tarp-covered boat in a suburban Boston backyard. He is in a federal prison and faces a charge of using a weapon of mass destruction to kill.

The Tsarnaev brothers' mother insists the allegations against them are lies.

Three of Dzhokhar Tsarnaev's friends, college classmates, were arrested and were accused of helping after the marathon bombing to remove a laptop and backpack from his dormitory room before the FBI searched it.

A top Republican senator on Thursday asked President Barack Obama's administration to explain how one of the students entered the United States without a valid student visa.

Sen. Chuck Grassley, of Iowa, in a three-page letter to Department of Homeland Security Secretary Janet Napolitano, asked for additional details about the student visa applications for Azamat Tazhayakov and Dias Kadyrbayev, college roommates from Kazakhstan charged with obstruction of justice in the marathon bombing case, and how Tazhayakov was allowed to re-enter the United States in January.

Tazhayakov was a student at the University of Massachusetts Dartmouth when he left the country in December. In early January, his student visa status was terminated because he was academically dismissed by the university.

The third student arrested, Robel Phillipos, was charged with willfully making materially false statements to federal law enforcement officials during a terrorism investigation.

The lawyers for the Kazakh students said their clients had nothing to do with the bombing and were just as shocked by it as everyone else. Phillipos' attorney said the only allegation against him was "he made a misrepresentation."

Scientists revolutionize creation of genetically altered mice to model human disease

May 2, 2013 ? Whitehead Institute Founding Member Rudolf Jaenisch, who helped transform the study of genetics by creating the first transgenic mouse in 1974, is again revolutionizing how genetically altered animal models are created and perhaps even redefining what species may serve as models.

"This new method is a game changer," says Jaenisch, who is also a professor of biology at MIT. "We can now make a mouse with five mutations in just three to four weeks, whereas the conventional way would take three to four years. And it's rather straightforward, probably even easier than the conventional way."

Scientists create models in mice by altering specific genes that have been associated with a given disease. The models allow for the study of the development and course of the disease and the effects of various interventions, including genetic and chemical. For the past 20 years, the creation of such models has remained relatively unchanged: scientists insert a piece of DNA into a mouse embryonic stem (ES) cell, inject the modified cell into a very early-stage embryo, called a blastocyst, then implant this developing ball of cells into a foster female mouse. The whole process can take years and tens of thousands of dollars to establish a mouse strain with, for example, a single copy of a gene "knocked out." Such knockouts can only be created in very few species, including mice and rats, whose ES cells can be grown and modified reliably.

The new approach used by scientists in Jaenisch's lab bypasses ES cells to quickly and efficiently produce mice with mutations in both copies of multiple genes. In next week's issue of the journal Cell, Haoyi Wang, Hui Yang, and Chikdu Shivalila describe their technique, which is based on a system that certain bacteria use to fend off viral attacks.

This is the first time that the system, known as CRISPR (for "clustered regularly interspaced short palindromic repeat")/Cas (for "CRISPR-associated"), has been used to alter multiple genes in a single multicellular organism. Shivalila says the process is so accessible that he expects other labs to adopt it quickly.

"For any institution or university with a core facility, we think this will be the way they will start making mice carrying specific mutations because it's a lot faster and so efficient," says Shivalila, one of Jaenisch's graduate students. "We were surprised that we could get two genes 'knocked out' at four loci very, very efficiently, about 80% efficiency. If we used TALENs, a more recent and complicated development in genetic engineering, we got 30% efficiency for just one gene."

Because the CRISPR/Cas technique can generate mutant mice even without using ES cells, a limitation of the conventional method for making models, genetic research may no longer be confined to a limited list of model organisms -- those for which ES cells exist.

"This breaks down the definition of model organism," says Wang, a postdoctoral researcher in Jaenisch's lab. "So now, even with limited resources, any animal with established embryo manipulation procedures could be the subject of genome engineering. With many of the animals' genomes that have been sequenced, we could use this technology to establish efficient genetic manipulations in more species, to study the unique biology of each, and to learn more about evolution."

Thus, Wang, Yang, and Shivalila have used CRISPR/Cas to create mouse models only, but the team is excited broaden its application to other animals.

"We also need to see if the CRISPR/Cas system has any unexpected, undesired off-target effects, changes to the genome that we don't want," says Yang, a postdoctoral researcher in the Jaenisch lab. "So we need study this further to establish the fidelity of the system. But I think this will be the way to go."

Ashton Kutcher's A-Grade Fund Raising At $100 Million Valuation

IMG_8321Today at TechCrunch Disrupt NY, Ashton Kutcher took the stage with Guy Oseary to talk about their A-Grade fund. Arrington started right away about a rumor that the fund is raising money at a $100 million valuation. The two partners confirm the rumors, saying that they are raising “enough money.” It means that financial institutions and companies will invest a certain amount of money in exchange for equity in the A-Grade fund, valuing the existing investments and activities at $100 million. With three partners (Ashton Kutcher, Guy Oseary and Ron Burkle), the fund has been investing for about two and a half years. Until today, the three partners only invested personal money. It is still unclear how much money they put into the fund to date. Among its portfolio companies, the A-Grade fund has invested in Spotify, Uber, Shazam, Soundcloud, Fab and Airbnb. It mainly takes part in seed and Series A rounds. When asked whether it is bundling current investments or creating a new fund, Ashton Kutcher answered that it is doing both. “We’re just somewhat formalizing what we’ve been doing,” Kutcher said. The new fund will keep the A-Grade name. While the new funding is not official yet, Kutcher and Oseary said that it’s mostly a one-time investment. “We are pretty well filled up,” Kutcher said. The partners will make an announcement when the deal finalizes.
